I have the standard hot keys assigned for x,y&z.... But there are several options on the hot key screen that I don't know about...

Under the "System Hotkey" section, you can set hotkeys to do what the buttons say:

Select DRO (selects the last selected DRO)
Select MDI (will work the same as the "Enter" key)
Load G-code
Select the G-Code window

(My version of Mach3 has another option to come out of reset, you're version must be older)

OEM Triggers let you activate Mach3 functions from external buttons assigned to Ports and Pins > Input pins > OEMTriggers

Yes, but you'll need to edit the screen in a screen designer and write a macro to do it. I do something similar in my screenset.
